逆病毒介导EGFP基因转染人骨髓间充质干细胞的实验研究

摘要 目的探讨逆转录病毒介导增强型绿色荧光蛋白(EGFP)转染人骨髓间充质干细胞(hBM-MSC)的可行性。方法携带EGFP的重组逆转录病毒载体(EGFP-RV)转染体外培养扩增的hBM-MSC,嘌呤霉素筛选,获得表达EGFP的hBM-MSC/EGFP,以未转染的hBM-MSC为对照组,观察两组细胞的形态、生长特性、表面分子表达及向脂肪细胞、成骨细胞分化的潜能。结果EGFP-RV转染hBM-MSC后,获得稳定表达EGFP的hBM-MSC/EGFP;hBM-MSC/EGFP的细胞形态、生长特性、表面分子表达及向脂肪细胞、成骨细胞分化的潜能与未转染的hBM-MSC无差别。结论逆转录病毒介导的EGFP可作为人骨髓间充质干细胞的良好示踪剂。 Objective To explore the feasibility of transfection of human bone marrow mesenchymal stem cells (hBM-MSC) with retroviral vector containing enhanced green fluorescent protein (EGFP).Methods In vitro expanded hBM-MSC transfected with recombinant retroviral vetor containing EGFP and selected with puromycin. The morphology, cell growth, surface marker and the muhilineage differentiation to adipocyte and osteoblast were compared between transfected hBM-MSC (hBM-MSC/EGFP) and untransfected controls. Results hBM-MSC,/EGFP was established through transfection with retroviral vector. There were no significant difference in morphology, growth pattem, surface marker and multidifferentiation potential between the hBM-MSC/EGFP and non-transfected cells. Condusion The retrovirus mediated EGFP is a reliable tracer for hBM-MSC in vivo.
