摘要
目的为了进行肾移植的基础研究,建立一种可复制的大鼠原位异体肾移植模型。方法采用Wistar大鼠作为供体和受体,原位低温灌注、原位移植、显微外科技术肾动脉端端吻合、肾静脉cuff套管吻合、输尿管端端吻合重建尿路。结果经过近5个月的反复实验摸索,克服了移植物灌注、血管吻合、输尿管吻合等技术难关。建立了稳定可复制的实用的肾移植大鼠模型。模型成功率为86%,动脉吻合时间(14±2 min),肾静脉Cuff套管吻合时间(6±2min),供肾缺血时间小于40 min,受体存活可达2周以上。结论采用本方法制作的大鼠肾移植模型成功率高,手术方法简便可行,符合肾移植实验研究的需要,具有推广价值。
Objective Establishing stable rat orthotopie renal transplantation(ORT) model in order to investigate details of kidney transplantation. Methods Wistar rats act as both donators and receptors. Low temperature saline perfusion and transplantation of kidney in situ were performed.end-to-end anastomosis was used for renal artery connection, and the technique was also used in the connection of ureter in order to rebuilt urinary passage. The cuff technique was utilize for anastomosis of renal vein. Results Stable and pragmatic rat ORT model was established after overcoming technique difficuhies in grafts infusion, vascular and ureteric anastomosis for five monthes repeating experiments, achievement ratio was 86%, arterious anastomosis time is 14 ± 2 min., venous anastomosis time is 6 ± 2min. donor renal ischemia time lesses than 40 min., and the receptor rats are able to live more than two weeks. Conclusion The establishmentof the ORT model with reliable operational techniques had higher achievement ratio which could meet the need of other experiments in renal transplantations.
