摘要
目的基于雌性小鼠胚胎连续石蜡切片进行中肾旁管定位,探讨制备高质量的含中肾旁管的小鼠胚胎组织切片的方法。方法正常孕15.5 d、17.5 d、19.5 d小鼠的胚胎,聚合酶链式反应(PCR)鉴定性别,取雌性鼠胚石蜡包埋,连续横断切片,每4张取1张行HE染色后获取连续切片图像。根据雌性小鼠胚胎生殖管道解剖走形,识别并定位中肾旁管。结果获取含不同发育天数中肾旁管的小鼠胚胎切片。结论利用石蜡包埋连续切片对中肾旁管进行解剖定位,是有效获得含中肾旁管的小鼠胚胎石蜡切片的方法。
Objective To investigate a method of preparation of high quality paraffin sections of mouse embryonic tissue with paramesonephric duct based on the location of paramesonephric duct in paraffin block serial sections of mouse embryonic. Methods The sex of normal pregnancy 15.5 d, 17.5 d, 19.5 d mouse embryos was identified by polymerase chain reaction (PCR) ,and the female embryonic were embedded in paraffin block, and the specimens were sectioned serially and dyed of hematoxylin-eosine(HE) every four section. The paramesonephrie duct was located and recognized according to the reproductive tract anatomy of female mouse embryonic. Results The microtome section of embryonic with paramesonephric duct were obtained. Conclusion The paraffin embedding and serial sections can be used to locate the paramesonephric, and is an effective method to obtain paraffin section of mouse embryonic tissue with paramesonephric duct.
