摘要
目的研究组胺、右旋糖酐40和4-氨基吡啶(4-AP)3种致痒物质对瘙痒动物模型的影响,尝试建立一种新型的瘙痒动物模型。方法用正交实验方法安排实验,各组分别在脱毛部位皮下注射0.1 ml不同致痒剂,记录小鼠30 min内的搔抓潜伏期和搔抓次数,用综合评分法筛选最佳致痒组方。分别取各组小鼠注射部位皮肤,测定皮肤中的组胺含量。结果最佳造模组方为联合应用组胺和4-氨基吡啶;各试验组与空白组比较,30 min内的搔抓潜伏期和搔抓次数差异具有显著性(P<0.05或P<0.01);皮肤组胺含量差异亦具有显著性(P<0.05或P<0.01)。结论联合应用组胺和4-氨基吡啶可建立一种理想的瘙痒动物模型。
Aim To research the influence of the animals pruritus model by used pruritus medium with histamine, dextran 40 or 4-aminopyridine (4-AP), and to establish a new pruritus model. Methods The orthogonal test was used to array the experiment. In the experiment different pruritus mediums were hypodermically injected for 0. 1 ml in the depilated area, the scratching incubation period and scratching number in 30 minutes were counted after the injection. The best pruritus group was screened out by synthesis grading law. The injected skin area in each group was located out to be determined the histamine contents in it. Results The best model group was the combination of histamine and 4-aminopyridine. Compared with the blank group, the scratching number and scratching incubation period of other pruritus model groups in 30 minutes were of significant difference (P 〈 0. 05 or P 〈 0. 01 ) ;The histamine contents also were significantly different(P 〈 0. 05 or P 〈 0. 01 ). Conclusion An ideal pruritus model can be established by the combined use of histamine and 4-AP.
