摘要
目的探讨Exendin-4对高脂喂养的肥胖大鼠胰岛β细胞功能及糖脂代谢的影响。方法SD大鼠40只用简单随机分组法分为2组,分别给予普通饲料(NC组,10只)及高脂饲料(30只)。喂养10周后,高脂喂养大鼠分为高脂(HF)组,0.5、2μg/kg Exendin-4治疗高脂组(HFL、HFH组),每组10只。治疗6周后,用静脉葡萄糖耐量实验(IVGTT)和胰岛素耐量实验(ITT)来评价机体的胰岛β细胞功能及机体对胰岛素的敏感性,同时检测基础血脂水平。结果高脂诱导肥胖大鼠经Exendin-4处理后,Lee’s指数、空腹血浆游离脂肪酸(FFA)、甘油三酯(TG)、胆固醇(TC)明显降低(P<0.01);IVGTT和ITT指标明显改善(P<0.05),胰岛素分泌水平增高(P<0.01),胰岛β细胞功能改善(P<0.05),HFH组较HFL组上述指标改善更为明显。结论Exendin-4可改善肥胖大鼠的胰岛β细胞功能和糖脂代谢,并可能呈剂量-效应关系。
Objective To investigate the effects of Exendin-4 on islet β-cell function and glucose-lipid metabolism in obese rats induced by high fat diet. Methods Forty Sprague-Dawley (SD) rats were randomly divided into NC group (fed with normal chow diet, n = 10) and HF group ( fed high fat diet for 10 weeks, n = 30). The rats in HF group were subdivided into three groups according to the single dose of Exendin-4 (0, 0. 5, 2 μg/kg) that was intraperitoneally injected twice a day for 6 weeks. The isletβ-cell function, insulin sensitivity and glucose lipid metabolism in awake rats were evaluated by intravenous glucose tolerance test (IVGTT) and insulin tolerance test (ITT). Results Lee' s indexes, blood glucose (BG), plasma free fatty acids (FFA), triglyceride (TG) and total cholesterol (TC) levels were significantly increased in high fat diet rats, but they were significantly decreased after Exendin-4 treatment (all P 〈 0. 01 ). In these obese rats, Exendin-4 treatment achieved better results of IVGTT and ITT (P 〈 0. 05 ), also better islet β-cell function (P 〈 0. 05 ), and increased insulin secretion ( P 〈 0. 01 ), especially when a high dose of Exendin-4 was administered. Conclusion Exendin-4 treatment ameliorates islet β-cell function and improves glucose-lipid metabolism in obese rats induced bv high fnt diet , seemingly in a dose-dependent manner.
