摘要
目的:研究当归多糖(APS)对小鼠骨髓基质细胞(BMSCs)生长、粘附分子ICAM-1和VCAM-1表达及其对骨髓单个核细胞黏附功能的影响.方法:观察APS对小鼠BMSCs消化时间的影响;MTT法检测APS对小鼠BMSCs增殖及对骨髓单个核细胞黏附功能的影响;流式细胞术检测APS对BM-SCs表面ICAM-1和VCAM-1表达的影响.结果:100,200mg/LAPS使BMSCs达80%贴壁后消化时间明显缩短(P<0.05);50,100mg/LAPS组BMSCs增殖能力较对照组增强(P<0.05);200,300mg/LAPS能使BMSCs表面ICAM-1表达降低,而50,100,200,300mg/LAPS均可使VCAM-1表达降低(P<0.05);体内注射APS后BMSCs表面ICAM-1和VCAM-1表达均降低(P<0.05);100,200,300mg/LAPS使小鼠BM-SCs对单个核细胞黏附率减弱(P<0.05).结论:APS能促进小鼠BMSCs增殖,降低其表面黏附分子ICAM-1和VCAM-1的表达,进而使小鼠BMSCs达80%贴壁后消化时间明显缩短,对骨髓单个核细胞的黏附率减弱.
AIM: To explore the effects of angelica polysacchatides (APS) on the cell growth, the expression of intercellular adhesion molecule-1 ( ICAM-1 ) and vascular cell adhesion molecule -1 ( VCAM-1 ) in routine bone marrow stromal cells (BMSCs) and the adhesive rate of routine bone marrow mononuclear cells to BMSCs. METHODS : The effects of APS on the digestion time of the routine BMSCs were observed with inverted microscope. The proliferation of BMSCs and the binding capacity of murine bone marrow mononuclear cells to BMSCs were evaluated by MTT assay. The expressions of ICAM-1 and VCAM-1 in routine BMSCs were analyzed by flow cytometry. RESULTS:With 100,200 mg/L of APS, the digestion time of murine BMSCs obviously decreased compared with control group. The cultured ceils proliferated well in DMEM/F12 containing 50 and 100 mg/L APS. 200 and 300 mg/L of APS decreased the expression of ICAM-1. 50,100,200 and 300 mg/L of APS decreased the expression of VCAM-1. The expressions of ICAM-1 and VCAM-1 were also decreased by intraperitoneal injection of APS. With 100, 200 and 300 mg/L of APS, the adhesive capacity of BMSCs to murine bone marrow mononuclear ceils was inferior to control group. CONCLUSION: APS enhances the proliferation of BMSCs and decreases the expression of ICAM-1 and VCAM-1 in BMSCs, so as to shorten the digestion time of murine BMSCs, and decrease the adhesive rate of murine bone marrow mononuclear cells to BMSCs.
