摘要
研究了生长激素(STH)、胰岛素(insulin)单独或者联合作用对猪卵母细胞体外成熟及孤雌激活后卵裂的影响。结果表明:STH、胰岛素对卵母细胞体外成熟及孤雌激活后卵裂呈现双重效应。在mNCSU-23基础液中添加0.15μg/mLSTH组的成熟率和卵裂率显著高于对照组及0.01,0.05,0.1,2μg/mL组(P<0.05);添加5μg/mL,8μg/mL胰岛素组的成熟率显著高于对照组及0.5,2,10μg/mL组;添加5μg/mL胰岛素组的卵裂率也显著高于对照组及0.5,2,8,10μg/mL组;0.15μg/mLSTH与2μg/mL胰岛素联合添加组的成熟率和卵裂率最高,同其他组相比,差异显著。
In this study, the effects of STH and insulin on porcine in vitro oocyte maturation and cleavage after parthenogenetic activation were investigated. The results showed that STH and insulin had double effects on oocyte maturation and cleavage. The maturation and the cleavage rates of the oocytes maturated in mNCSU - 23 medium supplemented with 0.15 μg/mL STH were significantly higher than that of control and the other tested groups which supplemented with 0.01, 0.05,0. 1,2μg/mL STH(P〈0. 05). The maturation and cleavage rates of the oocytes maturated in medium supplemented with 5μg/mL insulin was also higher than other group. However, both of the maturation and cleavage rates of the oocytes maturated in medium supplemented with 2μg/mL insulin and 0. 15μg/mL STH was significantly higher than that of other groups (P〈0. 05), which represented the optimal condition for in vitro maturation of porcine oocvtes.
