摘要
目的探讨表没食子儿茶素没食子酸酯(EGCG)对高糖诱导的大鼠系膜细胞增殖及炎症因子(MCP-1和ICAM-1)分泌的影响。方法通过体外培养的大鼠系膜细胞进行研究。设立低糖对照组(NG)和高糖组(HG、E100、E200、E400,为分别含EGCG0、100、200、400μg.L-1),分别培养12、24、48h。CCK细胞计数法检测不同浓度EGCG作用下大鼠系膜细胞的OD值,判断细胞增殖情况。采用ELISA方法检测培养细胞上清中MCP-1和ICAM-1蛋白浓度。结果EGCG能抑制高糖时系膜细胞的增殖活性,其抑制作用呈剂量和时间依赖的方式。系膜细胞处于高糖环境时,MCP-1和ICAM-1蛋白表达上调,低浓度的EGCG对MCP-1和ICAM-1蛋白的抑制效果不明显,高浓度的EGCG能明显抑制MCP-1和ICAM-1的分泌。结论在体外高糖条件下,大鼠系膜细胞在48h内以增殖为主,一定浓度的EGCG可抑制其增殖。EGCG可在蛋白水平抑制高糖刺激的大鼠系膜细胞MCP-1和ICAM-1的分泌。
Objective To explore the effects of epigallocatechin-3-gallate (EGCG) on proliferation of mesangial ceils and the contents of Monocyte chemoattractant protein-1 ( MCP-1 ) and intercellular adhesion molecule-1 (ICAM-1) in high glucose cultured rat mesangial cells. Methods Rat mesangial cells were divided into control group (low glucose 5.5 mmol·L^-1 without EGCG) and experiment groups (high glucose 25 mmol·L^-1 with EGCG (0, 100,200,400μg·L^-1 ), each group was incubated for 12, 24, 48 hours. The growth inhibiting effect of EGCG on mesangial ceils was measured by CCK-8 methods. The levels of MCP-1 and ICAM-1 in supernatant were determined by enzyme-linked immunosorbant assay (ELISA). Results The results showed that hyperglycemia stimulated mesangial cell proliferation and release of MCP-1 and ICAM-1. EGCG could inhibit cell growth in a dose- and time-dependent manner. In addition, high concentrations of EGCG could also decrease the levels of MCP-1 and ICAM-1 in supernatant. Conclusion EGCG can significantly inhibit proliferation and secretion of MCP-1 and ICAM-1 in high glucose induced rat mesangial cells.
