摘要
比较香菇(Lentinula edodes)和黑木耳(Auricularia auricula)在不同水解条件下的单位质量吸光值.筛选适宜水解条件,并通过测定姬松茸(Agaricus blazei)、鸡腿菇(Coprinus comatus)和茶树菇(Agrocybe aegerita)3种食用菌的总糖含量,对筛选的水解条件进行验证;采用正交试验优化苯酚-硫酸法的反应条件;最后选用苯酚硫酸法和费林氏滴定法分别测定4种供试食用菌的总糖含量,并进行比较。试验研究表明。食用菌适宜水解条件为:30 mL盐酸,100℃水浴水解3 h;优化的苯酚-硫酸法反应条件为:5%苯酚1.5 mL.浓硫酸5.5 mL,反应时间15 min;苯酚硫酸法和费林氏滴定法两种方法测得的食用菌总糖含量无显著差异.因苯酚-硫酸法操作简单,且简化了前处理过程.故是国标法——费林氏滴定法较理想的替代方法之一。
Optimal conditions for the hydrolysis of Lentinula edodes and Auricularia auricula fruit body tissue and assay of the total sugar in the hydrolysates using the phenol-sulfuric acid method were determined using orthogonal testing. Optimal hydrolysis was achieved by adding 30 mL conc. HCI to 0.25 g tissue suspended in 100 mL distilled water and incubating at 100℃ in a water bath for 3 h. Optimal conditions for the phenolsulfuric acid assay were: 1.5mL 5%(w/v) phenol, 5.5 mL concentrated sulfuric acid, and 15 min reaction time. These optimized hydrolysis and assay conditions were confirmed by determining the total sugar content of fruit body tissue taken from Agaricus blazei, Coprinus cornatus and Agrocybe aegerita. Comparison of the phenol-sulfuric acid and Fehling's methods for total sugar determination of L. edodes, A. auricula, A. blazei and A. aegerita revealed that the phenol-sulfuric acid method was an ideal substitute for the Fehlingtitration method.
