不同频率电针耐受对κ阿片受体mRNA转录的影响 被引量:9

摘要 目的:研究2Hz及100Hz电针耐受对κ受体mRNA转录的影响。方法:每天给大鼠2Hz及100Hz电针1次,每次30min,连续电针6d,在电针的第1、3、6天分别取各个脑区及脊髓,提取RNA,然后用反转录聚合酶链反应(RTPCR)方法检测κ受体mRNA水平。结果:(1)电针后的第1天,各脑区及脊髓κ受体mRNA均无明显改变;(2)每天给大鼠针刺(不给电刺激)1次,连续多次针刺可使纹状体内κ受体mRNA显著增加,在针刺的第6天杏仁核内κ受体mRNA也轻度增加;(3)每天给大鼠100Hz电针1次,每次30min,连续电针3~6次可使脊髓中κ受体mRNA水平下降,同样现象也可在纹状体及杏仁核中观察到,2Hz电针的结果很不一致,它使PAG和纹状体内κ受体mRNA水平降低,而杏仁核和脊髓中的κ受体水平增强,或呈双相反应。结论:单纯针刺3~6d本身有一定的中枢效应,但是多次电针则在大多数情况下抑制κ受体mRNA的转录,提示内源性阿片肽大量释放也可引起阿片受体下调,这可能是电针耐受的机制之一。 Objective: To observe the effects of 2 Hz and 100 Hz electroacupuncture(EA) stimulation on the transcription of CNS κ opioid receptor mRNA. Methods: EA in 2 Hz or 100 Hz was administered 30 min once a day for 6 days, total RNA extracted from discrete brain regions and the spinal cord on the first, third, sixth days and the level of mRNA detected with the method of reversetranscriptionpolymerase chain reaction(RTPCR). Results: (1) No significant change of the abundance of κ receptor mRNA was observed in various brain areas and spinal cord 24 h after one session of EA stimulation. (2)In rats receiving acupuncture without electricstimulation once (30 min) every day for 6 days, the mRNA level of κ opioid receptor increased significantly in the striatum, as well as in amygdala on the 6th day. (3)A significant decrease of the κ receptor mRNA level was found after 3~6 sessions of 100 Hz EA stimulation in the spinal cord, striatum and amygdala. In contrast, changes induced by repeated 2 Hz EA stimulation were not consistent, κ receptor mRNA level decreased in PAG and striatum, increased or changed biphasically in the amygdala and spinal cord. Conclusion: (1) Insertion of the acupuncture needle in the acupiont without giving eletric stimulation might produce certain CNS effects. (2)In most cases, repeated EA stimulations inhibited the transcription of κ opioid receptor t hat may lead to the downregulation of κopioid receptor. This may serve as one of the molecular mechanisms for the development of EA tolerance.
