蛋白酶活化受体2对大鼠肝星状细胞分泌MCP-1的诱导作用 被引量:3

摘要 目的探讨蛋白酶活化受体2(PAR2)对大鼠肝星状细胞(HSC)分泌单核细胞趋化蛋白-1(MCP-1)的调控作用。方法选用培养7d的SD大鼠HSC并接种于培养板各孔内,分别用不同浓度PAR2激动剂、活性肽、反活性肽和抑制剂刺激,ELISA法检测培养上清MCP-1水平。结果PAR2激动剂和活性肽可引起HSCMCP-1释放增加,抑制剂可抑制HSC促MCP-1的释放作用,其反活性肽不引起MCP-1释放增加。结论在大鼠肝纤维化炎症反应中,PAR2可促进HSC分泌MCP-1,促进炎症反应,其反活性肽和抑制剂可能具有抗炎作用。 Objective To investigate the actions of protease-activated receptor 2(PAR2) on the secretion of monocyte chemoat tractant protein 1(MCP-1) from rat hepatic stellate cells(HSC). Methods HSC after 7d isolated and cultured from SD rats livers were transferred in a 24 well culture plate. The challenge was performed by addition of various concentrations of PAR2 agonist peptides SLIGRL and its reverse peptides LRGILS, trypsin and trypsin inhibitor named soybean trypsin inhibitor(SBTI) into each well respectively. After 2h,8h and 16h, the reactions were terminated by removal of the supernatant.from each well. A sandwich ELISA was used to determine the levels of MCP-1 in supernatants. Results SLIGRL could induce secretion of MCP-1, but the reverse PAR2 agonists had little effects on MCP-1 release. Trypsin inhibitor-SBTI could inhibit secretion of MCP-1 induced by trypsin. The time course showed that the actions of PAR2 agonist peptides SLIGRL and trypsin initiated at 2h and reached their peak at 16h. Conclusion PAR2 agonist peptides and trypsin are potent secretogogue of MCP-1 release from cultured HSCs from rat liver,and PAR2 antagonists and trypsin inhibitor may possess the ability to inhibit airway inflammation.
