重组蟹金属硫蛋白对长爪沙鼠脑缺血再灌注损伤的保护作用 被引量:2

摘要 目的研究重组蟹金属硫蛋白(recombinant metallothionein,rMT)对长爪沙鼠脑缺血再灌注损伤的的保护作用。方法将50~70g长爪沙鼠随机分为假手术组(Sham)、脑缺血再灌注组(I/R组)和MT预处理组(MT组),每组又分为4个亚组,采用双侧颈总动脉夹闭15min制作全脑缺血再灌注模型,分别再灌注2、3、5和7d,应用HE染色和TdT介导的原位末端标记(TUNEL)法标记断裂的DNA片段检测凋亡细胞观察脑组织变化。结果病理变化显示不同再灌注时间MT组的海马锥体细胞带增宽,细胞空泡化减少;脑缺血再灌注2d后,海马区的TUNEL阳性细胞增多,再灌注3d后,阳性细胞数最多,至再灌注5d、7d时,阳性细胞数逐渐减少。MT组的阳性细胞数在不同时间内均有所减少,与模型组有显著差异(P<0.05)。结论外源性给予MT可减少细胞凋亡的发生,减轻脑组织的损伤,对长爪沙鼠脑缺血疾病具有保护作用。 [Objective] To study protective effect of recombinant metallothionein of crab on cerebral ischemie reperfusion injury in gerbils. [Methods] 50-70 g gerbils were divided into three groups randomly: Sham group, cerebral ischemic reperfusion group (I/R) and MT administration group (MT). Each group were further divided into 4 subgroups (n =10). Transient cerebral ischemic reperfusion model was established by ligating bilateral carotid for 15 minutes. The pathological change of prosencephal tissue was observed by HE staining, the apeptosis of neuron was detected by the TdT-mediated dUTP-biotin nick end labeling (TUNEL) staining during reperfusion cerebral ischemic-reperfusion (2, 3, 5 and 7 d). The image analysis was performed. [Results] MT administration group was observed in light microscopic that the neuronal morphologic changes including cone cells strip of hippocampi area widen, vacuolar degemerate nerve cells reduced. The number of positive TUNEL in I/R group increased on the 2nd day after reperfusion, obviously increased on the 3rd day and decreased on the 5 td, 7 td day. The number of positive TUNEL in MT administration group are significantly fewer than I/R group (P 〈0.05). [ Conclusion] MT could relieve putrescence and cell apeptosis of neuron. MT might have a potential protection for the treatment of cerebral ischemic disease.
