摘要
目的探讨染料木素对中年、早期老年大鼠卵泡发育的影响。方法用染料木素对4和10个月龄大鼠进行灌胃[160mg/(kg.d)],连续用药4周后处死,分别取染料木素灌胃组(试验组)和对照组大鼠双侧卵巢,用苏木素-伊红(hematoxylin-eoxin,HE)染色观察不同发育阶段卵泡比例,同时对9~10个月龄大鼠行阴道涂片,观察其动情周期变化,连续6周。结果在染料木素灌胃组的4个月龄大鼠卵巢内原始卵泡、次级卵泡的比例升高,窦状卵泡、闭锁卵泡的比例下降;在染料木素灌胃组的10个月龄大鼠卵巢内原始卵泡、次级卵泡的比例升高,闭锁卵泡的比例下降,且灌胃组的10个月龄大鼠维持着规则的动情周期。结论染料木素能延迟中年、早期老年期大鼠卵巢内原始卵泡发育启动,抑制各级卵泡闭锁,而可能有益于延长卵巢的生殖寿命。
[Objective] To study the effect of genistein on ovarian follicle development in middle-aged and early old aged rats. [Method] 4- and 10-month-old rats were treated with genistein (160 mg/kg·d) by intragastrie administration for 4 weeks. Ovaries were collected from the rats of both experimental groups and control groups. The ratio of ovarian follicles at different development stages was examined by hematoxylin-eoxin staining. The estrous cycle patterns were determined by daily observations of vaginal smears for 6 weeks in 9-10-month-old rats. [Restilts] The percentage of primordial and secondary follicles increased, whereas, antral and atretic follicles decreased in the 4-month-old rat ovaries treated after genistein treatment. The percentage of primordial and secondary follicles also increased and atretic follicles decreased in the 10-month-old rat ovaries after genistein treatment. The 10- month-old rats treated with genistein maintained regular estrous cycles. [Conclusion] Our results suggests that genistein inhibit primordial follicle development and decrease follicles atresia. Genistein may benefit to prolong the reproductive life of oyary.
