表面活性剂强化提取黄姜中薯蓣皂苷的研究 被引量:21

Study on Intensification Effects of Surfactant on the Extraction of Dioscin from Dioscorea zingiberensis C.H. Wright

摘要 对7种表面活性剂强化提取薯蓣皂苷的效果进行了比较,筛选出对该提取过程具有较好强化作用的表面活性剂——十二烷基硫酸钠(SDS)。通过正交实验、单因素实验考察了提取温度、提取时间、w(SDS)、提取溶剂用量对薯蓣皂苷提取率的影响,获得了优化的提取条件:黄姜粉末5 g,提取温度70℃,提取溶剂用量12 mL/g黄姜,提取时间2 h,w(SDS)=3%,获得皂素92.59 mg。实验结果表明,SDS对水提取黄姜中薯蓣皂苷(7.74mg)有明显强化作用,提取效率高于传统的甲醇/水溶液(75.08 mg),接近索氏提取(95.67 mg)。 Seven surfactants were tested as intensifier of dioscin extraction from Dioscorea zingiberensis C. H. Wright. Sodium dodecyl sulfate (SDS) was screened out as the best intensifier. Effects of temperature,time,w(SDS) and amount of solvent on the yield of dioscin have been examined using orthogonal design and single-factor experiments. The optimum extraction conditions for 5 g Dioscorea zingiberensis C. H. Wright powder were found to be : temperature is equal to 70 ℃, time is equal to 2 h, w(SDS) is equal to 3% and amount of solvent is 12 mL per gram powder,the yield of diosgenin was 92.59 mg. Experimental results indicate that: SDS can intensify the extraction of dioscin remarkably, (7.74 mg for water without SDS). The extract efficiency of SDS aqueous solution is higher than that of the traditional methanol-water solution, a conventional solution for dioscin extraction (75.08 mg) and is close to the efficiency of soxhlet extraction. The new extraction method can greatly reduce the usage of harmful organic solvent (95.67 mg).
