摘要
目的:探讨不同剂量阿司匹林与黄芪颗粒合用后对阿尔茨海默病大鼠学习记忆能力的影响。方法:60只SD大鼠随机分为空白、假手术、模型、模型+阿司匹林3、10、30mg/kg+黄芪颗粒3g/kg6组,将β-淀粉样蛋白(Aβ1-40)10μg(1μl)注入大鼠双侧海马CA1区制AD模型,给药组造模前2小时灌服阿司匹林3、10、30mg/kg+黄芪颗粒3g/kg,其余组给予等量双蒸水,之后连续灌服2周,每天一次,第3周通过Morris水迷宫和穿梭箱实验观察大鼠记忆能力改变;之后处死大鼠,HE染色观察海马组织病理改变。结果:同模型组相比,阿司匹林3、10、30mg/kg+黄芪颗粒3g/kg三组寻找平台时间潜伏期明显缩短,末次跨平台次数增加,有统计学意义(P<0.05)。结论:阿司匹林与黄芪颗粒合用能够改善AD大鼠学习记忆能力,对AD有一定治疗作用。
Objective: To observe the effects of different dosage of Aspirine and Huangqi Grain on ameliorating the ability of learning and memory on the model rats with Alzheimer's disease (AD).Methods: Sixty rats were randomly divided into control,sham operation,model and model + aspirine 3,10,30mg/kg +Huangqi Grain 3g/kg groups; the AD model rats were established by injecting Aβ1-40 10ug into both CA1 districts of hippocampuses; the treatment groups were adminstrated proper dosge of aspirine and Huangqi Grain 2h before operation, and continued,once a day for 2 weeks;the other groups were adminstrated equal dosage of double distilled water.The changes of learning and memory ability of rats were observed by the morris water maze and shuttle box test,and the change of pathology in the hippoeampus were observed by hematoxylin-eosin stain(HE). Results: Compared with sham operation group,the model group rats had extended escape latency , reduced crossing platform times (P〈0.05). Compared with model group, the there treatment groups shortened the escape latency and, improved crossing platform times (P〈0.05). Conclusion: The combined use of aspirine and Huangqi grain could ameliorate the ability of memory and learning.
