摘要
近年来,环介导等温扩增技术已经被开发利用。它采用能特异识别靶序列上6个位点的4条引物及一种具有链置换活性的DNA聚合酶,在恒温条件(60℃~65℃)下,不到1h的时间里进行核酸扩增,其扩增效率可达到10^9~10^10个数量级,具有特异性强、等温灵敏、操作简单、产物易检测等优点。环介导等温扩增法已经被用来快速检测动物病原(病毒、细菌、寄生虫)。论文就环介导等温扩增法的原理、特点及其在动物病原快速检测中的应用等做了简要的综述。
In recent years,a novel nucleic acid amplification method at a constant temperature, termed loopmediated isothermal amplification (LAMP) has been developed and used. This method employs a strand displacement activity DNA polymerase and a set of four specially designed primers that recognize a total of six distinct sequences on the target DNA. The cycling reaction continues with accumulation of 10^9-10^10 copies of target at a constant temperature of 60-65 ℃ in less than an hour. The LAMP assay has been adopted widely for the detection of animal pathogens(viruses,bacteria, parasites), because of its simplicity, high sensitivity and specificity and detecting easily products etc. The summary demonstrated the principle, characteristics and application in detecting rapidly animal pathogens of the LAMP assay and so on.
