山茛菪碱对大鼠供肝热缺血再灌注损伤的保护作用 被引量:1

The protective effect of hyoscyamine preconditioning on warm ischemia-reperfusion injury of donated liver in rats

摘要 目的探讨山莨菪碱预处理对无心跳大鼠供肝热缺血再灌注损伤的保护作用。方法根据山莨菪碱预处理与否及供肝获取前经历的供体心脏停搏时间30min或45min,将实验动物分为4组,即非预处理的30min(N30)组和45min(N45)组,山莨菪碱预处理的30min(tN30)组和45min(tN45)组行原位肝移植。观察存活状况,并于移植术后1d、3d、7d采集血样检测肝功能。结果N30组和N45组的1周存活率分别为50.8%和14.9%,而tN30组和tN45组的1周存活率分别为86.6%和65.8%,预处理组移植肝脏功能状态明显好于非预处理组。结论山莨菪碱预处理能够减轻供肝的热缺血再灌注损伤,改善肝功能,提高大鼠肝移植的存活率。 Objective To explore the protective effect of Hyoscyamine preconditioning on warm ischemia -repufusion injury (WIRI) of the liver from nor - heart beating donor(NHBD) in rats. Methods According to pretreatment with Hyoscyamine or not and the nor - heart - beating time of 30 or 45 min before donor liver was harvested , they were divided into 4 groups : the group of nor - heart - beating for 30 min( N30), the group of nor-heart - beating for 45 min ( N45 ) , the group of treatment and nor - heart - beating for 30 minutes( tN30), and the group of treat- ment and nor - heart - beating for 45 rain(tN45). Liver transplantation was perfonned respectively. The survival condition was observed. Plasma AST, ALT and LDH at 1 ,3 and 7 day(s) after transplantation were assayed. Results One - week survival rate in N30 and N45 groups was 50.% and 14.9% respectively, and one - week survival rate in tN30 and tN45 group was 86.6% and 65.8% respectively. Conclusion Hyoscyamine preconditioning can relief warm ischemia reperfusion injury of the donated liver, improve the graft liver function and enhance survival rate of rats bearing liver transplantation.
