摘要
[目的]研究菊芋中菊糖的提取分离方法。[方法]菊芋经预处理后粉碎,得到粗菊粉,粗菊粉浸提后,经石灰乳除杂、Sevag法脱蛋白、活性炭脱色、乙醇沉淀等过程进行纯化得到菊糖,测定了总糖、还原糖的含量。[结果]TS4厂家生产的粉末状活性炭对粗菊粉浸提液的脱色效果最好。经纯化处理后,利用热水浸提法得到的菊芋菊糖粗提取液,清澈透明。纯化液经浓缩、乙醇沉淀后,得到黄色的精制菊糖,菊糖的含量为20.23%,纯度为92.60%。[结论]菊芋中菊糖的最佳提取方法为采用热水浸提法,即以固液比1∶15,80℃,浸提90min;再通过石灰乳除杂、Sevag法脱蛋白、活性炭脱色、乙醇沉淀等过程对粗提取液进行纯化。
[Objective] The aim was to study the extraction and isolation method of inulin from Jerusalem artichoke.[Method] After pretreatment,J.artichoke was grinded to obtain the crude inulin.Through impurity removal by lime milk,protein removal with Sevag method,decoloration with active carbon and ethanol precipitation,the crude inulin was extracted and purified to get the refined inulin.The content of total sugar and reducing sugar was measured.[Result] The decoloration effect of powdery active carbon produced by TS4 manufacturer on the crude inulin extract was the best.After purification,the crude inulin of J.artichoke obtained by hot water extraction method was clear and transparent.After the purification solution was concentrated and precipitated with ethanol,the yellow purified inulin was obtained.Its inulin content was 20.23% and its purity was 92.60%.[Conclusion] The optimum extraction method of inulin from J.artichoke was that hot water extraction method was taken with solid-liquid ratio of 1∶15,extraction for 90 min at 80 ℃,and through the impurity removal by lime milk,protein removal with Sevag method and decoloration with active carbon,the crude inulin could be purified by ethanol precipitation.
