摘要
目的:优化样品前处理条件和色谱条件,建立一种固相萃取-高效液相色谱法测定食品中丙烯酰胺含量。方法:样品中丙烯酰胺经水溶解、超声提取、正己烷脱脂、Carrez试剂沉淀蛋白,采用活性炭固相萃取柱净化后进行HPLC分析,流动相为甲醇:水(10:90,v/v),流速为0.8 ml/min,进样量为10μl,检测波长为205 nm。结果:该方法的线性范围为10~1000μg/L,r=0.9992,检出限为12μg/kg。薯片样品中丙烯酰胺的加标回收率为67.83%~82.05%,相对标准偏差为6.99%。结论:该方法经济、简便、灵敏、准确,可向常规实验室推广进行批量检测。
Objective:To establish a solid phase extraction -high performance liquid chromatography method for the determi- nation of acrylamide in foods by optimizing sample treatment and chromatography condition. Methods:Acrylamide in samples was dissolved with water, extracted with ultrasonic wave, defatted with n -hexane, cleaned by Carrez solution, purified by activated carbon solid phase extraction cartridges and then analyzed by HPLC. The mobile phase was 10% methanol water solution, the flow rate was 0. 8 ml/min, the volume of injection was 10 μl and the detection wavelength was 205 nm. Results:The lineafity range of the method was 10 - 1000 μg/L with correlation coefficient of 0. 9992. The detection limit of the method was 12μg/kg. The recov- eries ranged from 67.83% - 82. 05%, and the relative standard derivation was 6.99%. Conclusion:This method is cheap, con- venient, sensitive and accurate and it can be applied in general laboratories to determine acrylamide in batches.
