从大豆胚芽中分离纯化大豆皂苷的研究 被引量:5

摘要 以大豆胚芽为原料,通过溶剂萃取,大孔吸附树脂与阳离子交换树脂除杂处理,凝胶渗透色谱初步纯化,然后利用高效液相色谱纯化制备了5个大豆皂苷单体,液相色谱-质谱联用鉴定结果为两个A组大豆皂苷Aa、Ab,三个B组大豆皂苷Ba、Bb和Be。通过蒸发光散射与紫外检测并用,高效液相色谱分析其纯度皆达到95%以上,可作为标准品用于大豆皂苷含量的分析检测。 After ethanol extraction from soybean hypocotyls and removing impurities by macroporous absorbent resins and cation exchange resins, the extracts were applied on gel filtration for primary purification of soya saponins. Five pure soya saponin components were obtained by preparative HPLC from the extracts. The prepared soya saponins were elucidated by LC/MS as two saponins of group A, Aa and Ab, three saponins of group B, Ba, Bb and Be, respectively. The prepared soya saponins were also analyzed by HPLC coupled with evaporative light scattering detector and photodiode array detector. As a result, the purity of the prepared soya saponin components are more than 95% and can be employed as authentic reference for soya saponin content determination concerning soybean or related products.
