摘要 目的探讨表没食子儿茶素没食子酸酯(EGCG)对人结肠癌细胞株HT-29基质金属蛋白酶-9(MMP-9)表达的影响。方法应用RT-PCR法和Western blot法检测EGCG干预前后HT-29结肠癌细胞中MMP-9 mRNA及MMP-9蛋白表达。结果EGCG25、50、100μg/ml作用于结肠癌细胞株HT-2948h后,MMP-9 mRNA及MMP-9蛋白的表达量随着EGCG浓度的增加而下降,结果有统计学差异(P<0.05)。结论EGCG可显著抑制HT-29细胞中MMP-9 mRNA及MMP-9蛋白的表达,呈剂量依赖性。 Objective To study the effect of Epigallocatechin-3-gallate(EGCG) on matrix metalloproteinase 9(MMP-9) in human colon cancer cell line HT-29 in vitro. Methods In vitro, the MMP-9 mRNA and protein expression in human colon cancer cell line HT-29 was analyzed by RT-PCR and Western blot. Results In vitro, human colon cancer cell line HT-29 was treated by EGCG in various concentrations( 25, 50, 100μg/ml) . After 48 hours, the expression of MMP-9 mRNA and protein in human colon cancer cell line HT- 29 were gradually decreased with the increasing dose of EGCG( P 〈 0.05). Conclusion EGCG could inhibit the expression of MMP-9 in HT-29 cell in vitro and in dose dependent manner, and maybe it is one of the mechanizms of the anti-tumor effect of EGCG.
