嗅鞘细胞移植后脊髓半横断大鼠睫状神经营养因子和FGF-2表达的变化 被引量:3

Changes on CNTF and FGF-2 expression in rats with spinal cord semisection following olfactory ensheathing cell transplantation

摘要 目的:研究嗅鞘细胞(olfactory ensheathing cells,OECs)移植到损伤的脊髓后,能否分泌睫状神经营养因子(CNTF)和碱性成纤维细胞生长因子(FGF-2),以及能否引起脊髓组织内CNTF和FGF-2表达的变化及脊髓的细胞有何数量改变。方法:体外分离培养大鼠嗅球嗅鞘细胞,通过免疫荧光细胞化学染色检测其CNTF和FGF-2的表达。将嗅鞘细胞移植入脊髓半横断SD大鼠,观察术后7天和21天嗅鞘细胞在脊髓内的存活情况,并对脊髓组织切片进行CNTF和FGF-2免疫荧光组织化学染色,计数阳性细胞并计算阳性面积百分比。结果:嗅鞘细胞在体外培养和移植后都能表达CNTF和FGF-2。同未移植组对比,移植组脊髓CNTF和FGF-2表达明显增强(P<0.05),神经元和少突胶质细胞数量增加(P<0.05),星型胶质细胞数量无明显改变。结论:嗅鞘细胞移植入损伤的脊髓后能分泌CNTF和FGF-2,并使脊髓组织内CNTF和FGF-2的表达增加,引起神经元和少突胶质细胞数量增加。 Objective: To investigate whether olfactory ensheathing cells(OECs) could secret ciliary neurotrophic factor(CNTF) and basic fibroblastic growth factor(FGF-2),and make expression change of them when planted in the spinal cord,then count the change of number for spinal cord cells.Methods: To culture and identify the rat OECs.We detected the expression of CNTF and FGF-2 by immunofluorescence cytochemistry in OECs cells.OECs were transplanted into the hemisectioned spinal cord of adult SD rats and their survival condition was observed at 7th days and 21st days after transplantation.The CNTF and FGF-2 positive cell number and the positive area percentage,as well as the number of neuron and neuroglia calculated by immunofluorescence analysis in the spinal cord. Results: OECs could express CNTF and FGF-2 in vitro and in viro post transplantation.Significantly higher levels of CNTF and FGF2 expression were detected compared with control(medium-injected)rats(P〈0.05).The number of neuron and oligodendrocyte increased dramatically surrounding the wound(P〈0.05),but not astrocyte. Conclusion: Post-transplanlation,OECs secreted CNTF and FGF2,and increased expression of CNTF and FGF-2 and the number of neuron and oligodendrayte in the spiual cord.
