血必净对大鼠外伤性脑损伤后血清S100B和IL-6水平的影响 被引量:21

摘要 目的:探讨血必净对大鼠外伤性脑损伤的神经保护机制。方法:将40只健康成年雄性SD大鼠随机分成4组:正常对照组、血必净治疗组、胞二磷胆碱治疗组和生理盐水组。采用改进的Feeney氏法制作大鼠自由落体脑创伤模型。伤后第6小时、第24小时、第3天、第5天、第7天断尾采血,运用双抗体夹心ELISA方法检测各组治疗后血清中S100B蛋白和白细胞介素-6(interleuk in-6,IL-6)的含量。结果:与正常对照组相比,血必净治疗组、胞二磷胆碱治疗组和生理盐水组血清中S100B蛋白和IL-6的含量均显著升高(P<0.001);与生理盐水组相比,血必净治疗组和胞二磷胆碱治疗组血清中S100B蛋白和IL-6的含量显著降低(P<0.001);与胞二磷胆碱治疗组相比,血必净治疗组血清中S100B蛋白和IL-6的含量显著降低(P<0.05)。结论:血必净可能通过抑制脑外伤后S100B和IL-6的合成与分泌,降低全身和局部的炎症反应程度,从而发挥神经保护作用。 Objective: To explore the neuroprotective mechanisms of Xuebijing on rats after traumatic brain injury.Methods: Forty healthy adult male Sprague-Dawley rats were randomly devided into 4 groups: normal control,Xuebijing,nicholin and saline groups.The model was made by improved Feeney′s traumatic brain injury model.Serum samples were collected at 6h,24h,3d,5d,7d after trauma.The serum S100B protein and IL-6 content of each group after treatment was detected with double antibody Sandwich ELISA. Results: Compared with the normal control group,the serum S100B protein and IL-6 content significant difference in Xuebijing,nicholin and saline groups(P〈0.001);compared with the saline group,the serum S100B protein and IL-6 content significantly lower in Xuebijing and nicholin groups(P〈0.001);compared with the nicholin group,the serum S100B protein and IL-6 content significantly lower in Xuebijing group(P〈0.05). Conclusion: Inhibition of the composition and secretion of S100B protein and IL-6 by Xuebijing might be one of the mechanisms in decreasing inflammatory reaction systemic and local,and this may be how Xuebijing exerts its neuroprotective effect on traumatic brain injury.
