摘要
[目的]采用不同诱变方法对兽疫链球菌(Streptococcus zooepidem icus)进行处理,以期达到提高发酵法生产透明质酸产量的目的。[方法]具体讨论了紫外、DES、微波、温度等不同诱变方法单独和复合诱变对兽疫链球菌产透明质酸的影响。[结果]复合诱变比单一诱变效果好。而最优的复合诱变处理方式为首先紫外诱变处理90 s,然后微波诱变处理100 s。在此条件下得到的菌株,其透明质酸产量由原始菌株的1.99 g/L提高到3.42 g/L,相对于原始菌株增加了72%,极大地提高了透明质酸产率。
[Objective] This study aimed to increase the output of hyaluronie acid by fermentation in the way of mutating Streptococcus zooepiclemicus. [Method] Streptococcus zooepidemicus was mutated by UV, DES, microwave and temperature respectively and the combination of them to compare their hyaluronic acid production. [Result] The results showed that the effect of combined mutation method on hyaluronic acid production was obvious than that of single method. And the best combined mutation method was that mutated by UV for 90 s firstly, then by microwave for 100 s, by which the yield of HA could be increased from 1.99 g/L to 3.42 g/L, and 72% higher than the initial yield, in this way the production of HA was improved greatly.
