摘要
目的考察胺碘酮对美托洛尔药代动力学的影响。方法将符合入选条件的病例分为2组,每组10例,分别口服美托洛尔、美托洛尔+胺碘酮,采用HPLC法测定美托洛尔血药浓度,经DAS软件处理,计算美托洛尔的主要药代动力学参数。结果合用胺碘酮后,美托洛尔的血药浓度和药时曲线下面积(Area Under theCurve,AUC)显著升高。结论胺碘酮可升高美托洛尔在人体内的血药浓度,导致发生不良反应的风险增加。
Objective To study the effect of amiodarone on metoprelol pharmacokineties. Methods Patients of two groups was selected （ 10 cases for each group） ,and they were given oral metoprolol or combination of metoprolol with amiodarone. The plasma concentration of metoprolol was assayed by HPLC, and the main pharmacokinetic pararneters were calculated with the DAS software. Results The combination of amiodarene,metoprolol plasma concentration and the AUC （ Area Under the Curve, AUC） were significantly increased. Conclusion Amiodarone can increased the metoprolol plasma concentration, and lead to increase of the adverse reactions risk.
