摘要
采用正交试验设计方法,对ISSR的影响因素进行研究,从Taq酶、Mg2+、dNTP和引物4因素3水平进行优化试验,筛选并建立了适合棉花黄萎病菌的ISSR-PCR最佳反应体系,即25μL的反应体系中含有1×PCR buff-er,200μmol/L dNTP,0.75μmol/L引物,150 mmol/L Mg2+,1U TaqDNA聚合酶和40 ng模板DNA。
In this study, the orthogonal design was used to optimize ISSR amplification system on Verticillium dahliae of cotton in four factors (Taq poly-merase,Mg^2+ , dNTP, primer) at three levels respectively. A suitable ISSR- PCR reaction system was established ,namely 25 μL reaction system containing 1 × PCR buffer, 200 μmol/L dNTP,0. 75 μmol/L primer , 150 mmol/L Mg^2+ , 1UTaq DNA po- ly-merase and 40ng DNA templet.
