拮抗TMV菌株By33产抗病毒蛋白分离纯化及发酵特性研究 被引量:2

摘要 从烟田耕层土壤分离获得对TMV有拮抗作用的菌株By33。其无菌发酵液对TMV的体外钝化作用为81.81%,涂抹菌液24h后接种,仍有73.01%的防效;硫酸铵沉淀获得的活性粗蛋白对TMV体外钝化作用为85.35%。粗提蛋白液经PEG浓缩、凝胶过滤和柱层析分离纯化,再经SDS-PAGE电泳确定该蛋白分子量为27.5KDa,对TMV的钝化效果为80.8%。NB培养基在初始pH6.5、温度30℃、通气量150mL/500mL、180r/min发酵培养48h,产物活性最高。 Research on antagonistic bacteria named By33 isolated from soil of tobacco field was conducted. Results showed that the mixture of unfermentative liquids of By33 with TMV had the passivation effect of 81.81 % . The prevention control was 73.01% when TMV was inoculated 24 hours after spraying the fermentative liquids of By33. The antivirus protein obtained by ammonium sulfate precipitation had the effect of 85.38% to TMV. The molecular weight of the active protein was 27.5KDa after being concentrated with PEG and purified with gelatine filtration and SDS-PAGE, which had the passivation effect of 80.8% to TMV. The optimum culture media was NB with initial pH 6.5. Culturing for 48h at 30℃ and supplying oxygen as much as 150 mL/500 mL was good at fermentation of bacterial strain.
