摘要
研究了窖泥放线菌的分离方法,结果表明,新鲜窖泥制成10%窖泥悬浮液,加0.5g/kg的SDS和10g/kg酪蛋白水解物、40℃振荡20min、稀释10倍后在含有15mL/L黄水和5mg/L重铬酸钾的高氏I号分离平板最有利于窖泥放线菌的分离。同时,经过优化分离,发现新鲜窖泥中放线菌数量为9.6×104个/g,并且菌落特征和细胞表观特征较为丰富。
The optimum isolation method of actinomycetes from pit mud was studied. The results showed that the isolation processes were as follows: after the collected fresh pit mud suspension (10 %)added with SDS (0.5 g/kg ), casein hydrolysate(10 g/kg) was stirred at 40℃ for 20 min, then the pit mud suspension diluted with water to l0 % and then inoculated with the reformative Gaoshi No.1(added 15 mL/L yellow water and 75 mg/L K2Cr2O7) for the isolation. The number of actinomycetes in pit mud reached 9.59×10^4 per gram and the shapes of colons and cells showed rich diversity.
