摘要 考察添加剂对绿色木霉产纤维素酶的影响。结果表明,初始培养基中添加油酸、十二烷基磺酸钠(SDS)、麦芽糖、羧甲基纤维素(CMC)、水杨素和水解淀粉等对绿色木霉产纤维素酶有促进作用,其中添加2%油酸和1%SDS均可使酶活提高1倍以上。在木质纤维素酶的水解过程中,在酶液中添加Mg2+、K+、Co2+、Tween80、聚乙二醇(PEG)等均对酶有激活作用;当底物浓度为30%,酶量1.0FPA/mL,PEG6000浓度0.14%,Co2+浓度为0.06%,搅拌速度120r/min条件下,能得到较高的糖浓度(85g/L)。乙醇发酵过程,发现木糖、葡萄糖单独发酵得酒精度分别为0.7g/100mL和4.3g/100mL;木糖-葡萄糖分步发酵的酒精浓度可达5.1g/100mL,而木糖-葡萄糖同步发酵得酒精浓度仅为4.4g/100mL。 The effects of the additives on cellulase production by trichoderma viride were investigated. The results showed that the addition of oleinic acid, sodium dodecyl sulphate (SDS), maltose, carboxymethyl cellulose (CMC), salicin and hydrolysed in primitive medium could promote the activity of trichoderma viride, and the addition of 2 % oleinic acid or 1 % SDS could double enzyme activity. During the hydrolysis of lignocellulose, the addition of metal ions such as Mg^2+, K^+, Co^2+ and surfactant Tween80, polyethylene glycol (PEG) etc. could activate enzyme activity. A higher sugar concentration (85 g/L) could be achieved when the concentration of substrate was 30 %, cellulase amount was 1.0 FPIU/mL, PEG4000 concentration was 0.14 %, Co2+ concentration was 0.06 %, and stirring speed was 120 r/min. During ethanol fermentation process, it was found that the alcoholicity by xylose fermentation or by glucose fermentation were 0.7 g/100 mL and 4.3 g/100 mL respectively, and the alcoholicity by xylose-glucose fermentation asychronously could reach up to 5.1 g/100 mL, while the alcoholicity by xylose-glucose fermentation simultaneously was only 4.4 g/100 mL.
