摘要
根据已发表的鸭圆环病毒(duck circovirus,DuCV)序列,合成了1对检测引物。利用此对引物对广东省新兴县发生的疑似圆环病毒感染的患病番鸭的肝、胰腺、心、肾、脾、肺、血液、腔上囊、胸腺、哈氏腺、粪便和羽毛进行了PCR扩增。结果显示,获得了与预期619 bp大小相符的DNA片段,经测序确认其为DuCV特异序列,证实鸭群感染了鸭圆环病毒。然后,再设计了2对引物从鸭组织提取的DNA中扩增出2条片段,拼接校对后获得了全基因组序列,并对其进行了分析。结果表明,在感染鸭体内各组织器官中都有DuCV存在,该株病毒基因组序列全长为1 988 bp,与部分已登录GenBank的DuCV同源性高达97.0%以上。
A pair of degenerated primers was synthesized based on the published duck circovirus (DuCV) sequences. A DNA fragment of 619 bp was amplified from extracts of the liver,spleen,kidney,fabricius, thymus, gland of harder, heart, blood, lung and pancreas of cireovirus-like infected ducks in Xinxing County of Guangdong Province using the primers,respectively. The fragment was then sequenced and anal- ysised,the result showed that these ducks were infected with DuCV indeed. Two nucleotide fragments (1 349 bp and 997 bp respectively) were amplified from DNA of various tissues of the infected ducks using another two pairs of degenerated primers. After they had been assembled together, the genome were obtained and analyzed. The results showed that DuCV could be detected in most of tissues of the infected ducks,the obtained sequence was a circular DNA with a total length of 1 988 nt,and the genome shared high identity of over 97.0% with some DuCV sequence available in GenBank.
