骨碎补水提液对大鼠成骨细胞的影响 被引量:14

摘要 为观察骨碎补水提液对体外培养大鼠成骨细胞增殖、分化的影响,用体外培养新生SD大鼠颅盖骨分离的成骨细胞,将不同浓度的骨碎补水提液分别与第3代大鼠成骨细胞进行体外共同培养,采用噻唑蓝(MTT)法测定细胞增殖能力,对硝基苯磷酸盐法(PNPP法)测定细胞的碱性磷酸酶(ALP)活性。结果显示,与对照组相比,作用48 h、72 h,骨碎补水提液均能显著促进大鼠成骨细胞的增殖,且与时效有关;作用48 h,10-3、10-2mg/mL骨碎补水提液浓度组可显著升高细胞ALP活性(P<0.05)。表明骨碎补水提液中存在较高活性的促大鼠成骨细胞增殖和分化的物质。 To investigate the effects of Rhizoma Drynariae aqueous-extracts on proliferation and differentiation of rat calvarial osteoblasts cultured in vitro. Osteoblasts were isolated from the skulls of 24 h newly born rats,the ceils of third passage were induced for screening potency of Rhizoma Drynariae aqueous-extracts. MTT was applied to determine proliferation of osteohlasts promoted by Rhizoma Drynariae aqueous-extracts at different dosages, differentiating effects with different concentrations in the cell were evaluated through the examinations of alkali phosphatase(ALP) activities by PNPP method. The results indi cated that the rat osteoblasts, which exposed Rhizoma Drynariae agueous-extraets, were proliferated significantly at 48 h and 72 h, ALP was also increased in the dosage of 10.3 mg/mL and 10^-2 mg/mL of Rhizoma Drynariae agueous-extracts at 48 h, It is time related to the proliferation. It was conoluded that the promoting contents of Rhizoma Drynariae agueous-extracts play an importawt role in the osteoblasts proliferation and differentiation in vitro.
