摘要
建立了动物组织(肌肉、肝脏、肾脏)中粘杆菌素、杆菌肽和维吉尼霉素等多肽类抗生素残留量的检测方法。样品经甲醇-0.1%甲酸提取,Oasis HLB固相萃取柱净化后,利用液相色谱-串联质谱进行定性、定量分析。3种多肽类抗生素的检出限分别为粘杆菌素25μg/kg,杆菌肽50μg/kg,维吉尼霉素20μg/kg;平均回收率分别为粘杆菌素89%~98%,杆菌肽90%~99%,维吉尼霉素92%~100%;相对标准偏差为1.91%~9.81%。方法具有快速简便、准确度和灵敏度高、重复性好等特点,适于动物组织中多肽类抗生素的测定。方法的技术指标满足国内外对多肽类抗生素残留量检测的要求。
A method was established to simultaneously detect colistin, bacitracin, virginiamycin residues in animal tissues, such as muscle, liver and kidney, by liquid chromatography tandem mass spectrometry. The samples were extracted by methanol -0. 1% formic acid, and cleaned up by Oasis HLB solid phase extraction column. The analytes were separated by liquid chromatography and deter- mined by liquid chromatography coupled to tandem mass spectrometry under electrospray ion and multiple reaction monitor(MRM) mode. The results indicated that the detection limits of colistin, baci- tracin and virginiamycin were 25, 50 and 20μg/kg, respectively, and their average recoveries were 89% - 98% , 90% - 99% and 92% - 100% , respectively. The relative standard deviations (RSDs) ranged from 1.91% to 9.81%. The method has the advantage of simplicity, sensitivity and repeatability, and is suitable for simultaneous determination of peptides antibiotics in animal tissues. The technical indicators of methods could meet the request of peptide antibiotic residue testing at home and abroad.
