摘要
目的研究红景天苷对局灶性脑缺血/再灌注(I/R)损伤后神经可塑性和DNA修复酶APE/Ref表达的影响。方法Wistar大鼠随机分为假手术组,I/R模型组和红景天苷干预组,采用线栓法制造大鼠大脑局灶缺血(2h)/再灌注模型,于灌注后0.25、1、3、7d4个时间点断头取脑,HE和尼氏染色观察神经元形态学变化,免疫组化方法检测无嘌呤无嘧啶位点/氧化还原因子-1(APE/Ref-1)的表达。结果与其他两组比较,红景天苷组梗死灶范围明显减小,梗死灶周围皮质神经元损伤明显减轻(P<0.05);APE/Ref-1阳性细胞数表达显著增高(P<0.05)。结论红景天可以易化脑I/R损伤后中枢神经系统神经再生和修复的可塑性,抑制APE/Ref-1降低可能是其在I/R早期发挥神经保护作用的一条重要途径。
Objective To study the effect of extract of salidroside on neuronal plasticity and the expression of apurinic apyrimidic site/redox factor (APE/Ref)-1 after focal cerebral ischemia/reperfusion (I/R) injury'. Methods The Wistar rats were divided into shamoperation, I/R model and salidroside groups. The model of middle cerebral artery occlusion 2 h/reperfusion ( MCAO 2 h/R) in rats was per-formed with the intraluminal filament occlusion. After 0. 25,1,3,7 d of reperfusion the rats were decapitated to obtain brain. The morphologi- cal changes were detected by HE stain and Nissl's staining. The APE/Ref-1 level was measured by immunohistochemical technology. Results After the pretreatment with salidroside ,the necrosis areas and the degree of pathological change in the peri-ischemic cortex neurons were obviously decreased, the number of APE/Ref-1 positive cells was significantly increased ( P 〈 0.05). Conclusions Salidroside can facilitate the neuroplasticity of regeneration and repairment in central nervous system after I/R injury. The activities of salidroside can be related to increasing APE/Ref-1 in cytoplasm, which would play an important role in nerve protection function in I/R.
