Notch signaling pathway and clinicopathology in human gastric cancer

摘要 目的探讨Notch信号通路在胃癌组织中的表达及意义。方法收集北京大学人民医院胃癌及正常胃黏膜组织标本63例,采用组织芯片免疫组化技术检测胃癌组织与正常胃黏膜中Notch信号通路相关蛋白的表达,分析这些蛋白与胃癌各临床病理指标之间的关系。结果本组63例胃癌组织中Notch信号相关蛋白Notch1、Jagged1、Delta1和NFκB的表达水平(分别为37%、29%、14%、27%)明显高于正常胃黏膜组织(分别为5%、2%、3%、2%)(P值均〈0.01)。胃癌组织中Notch1蛋白的表达与胃癌的分化程度有关(χ^2=3.944,P〈0.05)。胃癌组织中Jagged1蛋白的表达与NFκB的表达相关(r=0.486,P〈0.01)。结论Notch1信号通路过表达可能参与胃黏膜的癌变过程,而且Notch1的表达与胃癌恶性程度负相关,表明Notch信号通路在胃癌组织中的双重效应。 Objective To investigate Notch signaling pathway expression and its relationship with clinicopathological parameters in human gastric cancer. Methods Specimens of carcinoma tissues and normal gastric membrane were obtained from 63 gastric cancer patients undergoing radical surgery from Jan 1999 to Dec 2001. Notchl, Delta1, Jagged1 and NFκB protein were measured using tissure microarray and immunohistochemistry. Data were analysed using SPSS software. Results Notch1, Delta1, Jagged1 and NFκB protein were upregulated in gastric cancer tissue compared with adjacent normal tissue ( P 〈 0.01 ). The expression of Notchl protein in the well-differentiated tumors was higher than that in the poor- differentiated tumors ( P 〈 0. 05 ). Positive linear correlation was established between the expression of Jaggedl and NFκB. Conclusion Notchl signal transduction pathway plays an important role in the development of human gastric cancer by activation of NFκB through Notch1-Jagged1 signal transduction pathway. It is also evident that Notch signaling can be both oncogenic and tumor suppressive in human gastric cancer.
