摘要
目的探讨血管内皮生长因子(VEGF)反义寡核苷酸联合放射治疗对裸鼠移植瘤宫颈癌生长的抑制作用。方法制备Balc/C裸鼠皮下宫颈癌模型20只,随机分为4组:VEGF反义寡核苷酸(ASODN)治疗组、放射10Gy治疗组、VEGF反义寡核苷酸+放射10 Gy治疗组及对照组,每组5只。分别皮下注射ASODN,对照组注射生理盐水,每周2次,连续4周,放射剂量一次性给予。观察各组裸鼠肿瘤的生长情况、裸鼠的一般情况与生存期,测量肿瘤直径并计算瘤体积。断颈处死裸鼠,天平称量肿瘤的重量。免疫组化法测肿瘤VEGF。结果VEGF反义寡核苷酸和放射对肿瘤生长均有抑制作用,二者联合作用抑制更显著。放射使VEGF表达升高,ASODN使VEGF表达降低;肿瘤发生明显的凋亡形态学改变。结论反义寡核苷酸可以下调VEGF的表达,对放射治疗宫颈癌移植瘤有增敏作用,为宫颈癌的综合治疗提供了实验依据。
Objective To study the depressant effect of VEGF antisensenucleic acids united radiation therapy on nude mouse transplantation tumor uterine cervix cancer. Methods Twenty Bale/C athymic mice uterine cervix cancer analogues were divided randomly into 4 groups: VEGF ASODN group, radiation 10 Gy group, VEGF ASODN + radiation 10 Gy group and control group, 5 mice in each group. They hypodermic injected ASODN and normal saline to the control group, twice a week and 4 weeks totally. Radiation dose was given once. We observed the tumorous growth, general state of health and life span of all groups nude mice, measured tumor diameter and calculated tumor volume. The mice were sacrificed, their tumors were weighted by scalesl Then the tumor VEGF was measured by immunohistochemistry. Results VEGF antisense oligonucleotides and radiation showed depressant effect on tumor growth, the combination of them two had more notable effect. Radiation increased the VEGF expression and ASODN reduced the VEGF expression. Tumor had evident apoptosis morphology transform. Conclusion Antisense oligonucleotides can decrease VEGF expression, have the sensitization to the radiation therapy uterine cervix cancer transplantation tumor. It provides an experimental basis on the uterine cervix cancer combined therapy.
