TOLL样受体2和IL-2、6在梅毒患者外周血中的变化及其意义

Changes and clinical significance of toll-like receptor 2 in peripheral blood monocytes and levels of IL-2,6 in serum of symptomatic syphilitic patients

摘要 目的:观察梅毒患者外周血单核细胞Toll样受体2表达与血清中IL-2、6含量变化及其意义。方法:采用流式细胞仪检测60例显性梅毒患者外周血单核细胞表面TLR2蛋白表达水平(荧光强度)以及TLR2阳性细胞百分比,ELISA法检测血清IL-2、6水平,并以60例健康者作为对照组。结果:①显性梅毒患者外周血单核细胞TLR2的表达水平较对照组显著升高且一期低于二期梅毒患者,差异均有统计学意义(P<0.05);TLR2+CD14+细胞的阳性率低于正常对照组且一期低于二期梅毒患者,差异均有统计学意义(P<0.05)。②显性梅毒患者IL-2、IL-6均高于对照组,一期患者IL-2高于二期但IL-6低于二期,所有差异有统计学意义(P<0.01)。③TLR2的表达水平与TLR2+CD14+阳性率、IL-2无明显相关性,与IL-6呈显著正相关;IL-2与IL-6呈显著负相关。④显性梅毒患者外周血单核细胞TLR2的表达水平、TLR2+CD14+阳性率、IL-2、IL-6男女之间的表达无差异。结论:梅毒感染时TLR2、IL-2、IL-6显著增高且与病程有关,但与性别无关。 Objective:To observe the expression of Toll -like receptor 2 (TLR2) in peripheral blood monocytes and the changes of IL - 2, 6 in serum content of the symptomatic syphilitics. Methods: The protein expression of TLR2 ( mean fluorescence intensity, MFI ) and the precentage of TLR2 positive cells were evaluated through flow cytometric analysis with peripheral blood monocytes in 60 symptomatic syphilitics and 60 healthy controls. ELISA was used to detect the serum level of IL - 2, IL - 6. Results: ①The expression of toll - like receptor 2 in peripheral blood monocytes of symptomatic syphilitics was singnificantly higher than those in the controls ( P 〈 0. 05 ) , and TLR2 expressed on monocytes from primary syphilis was markedly lower than secondary syphilis (P 〈 0. 05 ) ;The precentage of TLR2 positive cell in peripheral blood monocytes of symptomatic syphilitics was singnificandy lower than those in the controls ( P 〈 0. 05 ), and TLR2 expressed on monocytes from primary syphilis patients was markedly lower than secondary syphilis( P 〈 0. 05 ). ②The serum level of IL- 2 and IL -6 were all ingnificantly higher than those in the controls(P 〈0. 05), The serum level of IL- 2 in primary syphilis was higher than that in secondary syphilisbut the IL- 6 was lower than in secondary syphilis ( P 〈 0. 05 ). ③The expression of toll - like receptor2 and the precentage of TLR2 positive cells had no significant correlation, nor the IL - 2, but positively correlated with the IL - 6. ④The expression of toll - like receptor2, the precentage of TLR2 positive cells and the changes of IL - 2, 6 in serum of the symptomatic syphilitics have nothing to do with gender. Conclusion :The expression of toll -like receptor 2 in peripheral blood monocytes and the serum level of IL -2 and IL -6 increased significantly when someone was infected Treponema gallidum and the course of disease, but has nothing to do with gender.
