水牛卵母细胞在不同培养液中成熟与凋亡的比较研究

摘要 为了解卵母细胞体外成熟与凋亡过程,进而提高卵母细胞体外成熟率,本试验研究了在培养液中添加不同浓度的表皮生长因子(EGF)、胰岛素样生长因子-1(IGF-1)对水牛卵母细胞体外成熟和凋亡的影响。结果表明:(1)添加各种浓度的EGF(10,20,30,50,100 ng/mL)均可以提高水牛卵母细胞的成熟率,降低卵母细胞的凋亡率,其中50 ng/mL EGF有显著影响(P<0.05);(2)添加各种浓度的IGF-1(10,30,50,100 ng/mL)均能提高水牛卵母细胞体外成熟率,降低卵母细胞的凋亡率,以30 ng/mL效果明显(P<0.05);(3)添加20 ng/mL EGF+30ng/mL IGF-1组卵母细胞体外成熟率和凋亡率分别高于和低于单独添加IGF-1和EGF组。 In order to study buffalo oocyte maturation and apeptosis in vitro, and to improve the increase rate of oocyte maturation, effects of EGF and IGF-1 on the maturation and apoptosis of buffalo oocytes were investigated in the present study. Addition of EGF at the different concentrations of 10, 20, 30, 50 and 100 ng/mL resulted in a respective decrease of apoptosis or increase of maturation in the proportion of buffalo oocytes, and addition of 50 ng/mL EGF into the maturation medium resulted in a significant effect. Addition of IGF-1 at the concen- trations of 10, 30, 50 and 100 ng/mL resulted in a respective decrease of apoptosis or increase of maturation in the proportion of buffalo oo- cytes, and addition of 30 ng/mL IGF-1 to the maturation medium resulted in a significant effect. More oocytes matured and few went apoptosis in the maturation medium containing 20 ng/mL EGF and 30 ug/mL IGF-1 in comparison with either 30 ng/mL IGF-1 or 20 ng/mL EGF group.
