肺心一号对实验性大鼠肺心病急性发作期血浆内皮素的影响 被引量:3

摘要 目的探讨肺心一号对野百合碱(MCT)诱发肺心病急性发作期的部分作用机制。方法将66只雄性W ister大鼠随机分为正常对照组、MCT组、硝苯吡啶治疗组及肺心一号低、中、高剂量治疗组,每组11只。分别于造模后14天灌胃相应药物,连续用药8天。采用改良右心导管术测定肺血流动力学参数,以非平衡法测定静脉血浆内皮素(ET-1)水平;处死大鼠后,称量肺湿重(wW)、右心室自由壁(RV)重和左心室加室间隔(LV+S)重,计算右心肥厚指数(RV/LV+S)。结果肺心一号和硝苯吡啶均能明显降低肺心病急性发作期大鼠的平均肺动脉压、wW、RV/LV+S及血浆ET-1水平(P<0.05,或P<0.01),以肺心一号高剂量组效果最好。结论肺心一号能有效降低肺动脉高压,改善心功能,其作用机制可能与显著降低血浆ET-1水平有关。 Objective To investigate the effect of Feixin ON-1 ( FXO-1 ) on experimental pulmonary heart disease in acute episode period(EPHDA) by means of measuring the level of plasma endothelin-1. Methods Adult male Wister rats were given a single dose of subcutaneous monocrotaline (50 mg/kg} to induce the model of EPHDA. A total of 66 rats were randomly divided into normal control group, EPHDA model group and Nifedipine group as well as low, moderate, or high dose of FXO-1 groups ( n = 11 ). At the 14th day after the model was established, gastric infusion of normal saline, FXM or Nifedipine were administrated respectively for 8 days. The parameters of pulmonary hemodynamics were monitored through the cannulation of polyvinyl tube. After rats were sacrificed, the wet weight of the lung, right ventricle and left ventricle added with interventricular septum were measured. The right ventricular thickening index was calculated. Results FXM and Nifedipine significantly inhibited the progression of pulmonary artery pressure, decreased the right heart index, the lung wet weight and the level of plasma endothelin-1 ( P 〈 0. 05 or P 〈 0. 01 ). Among them, the high-dose group got the best result. Conclusion FXM is able to reduce pulmonary hypertension and improve heart function. Its mechanisms may be related to reducing the level of plasma endothelin-1.
