摘要
目的:察通心络早期应用对糖尿病大鼠心肌病变的影响,探讨该药在防治糖尿病心肌病中的作用机制。方法:取36只大鼠,随机分为正常对照组、糖尿病组、通心络组。应用链腮佐菌素制备SD大鼠糖尿病模型。通心络干粉(1.43g生药/g干粉)0.3g·k^-1·d^-1。溶于水中灌胃。8周后检测各组大鼠血糖;测定左室收缩压和左室舒末压、左室内压最大上升和下降速率(-dp/dtmax);经衄染色,光镜下观察心肌病理变化;透射电镜观察心肌超微结构变化。结果:糖尿病组大鼠血糖明显升高。糖尿病组大鼠-dp,&max降低,而通心络组±dp/dtmax增高,差异具有显著性(P〈0.05)。光镜下糖尿病组心肌肌原纤维排列紊乱,肌丝成分减少;电镜下显示心肌内皮细胞明显肿胀,毛细血管基底膜出现不规则明显增厚。通心络组病变较轻。结论:通心络能明显改善糖尿病大鼠的心功能、能减轻糖尿病心肌病变;其作用机制可能是通过保护微血管来阻止糖尿病向糖尿病心肌病进展。
Objective :To observe the pathological changes ofdiabetic cardiomyopathy and apply Tongxinluo(TXL) on diabetesinduced cardiac disease,To mvestigate the mechanism ofdiabetic cardiomyopathy and protective role ofTXL on diabetic cardiomyopathy. Methods : Diabetic model was established by injection of streptozotocin (sTZ) in a dose of 60mg/kg 36 Sprague-Dawly rats were randomly divided into control group( CON), diabetic group (DM)and Tohgxiuluo treated diabetic rats (0.3g/kg · d drinked in the water aRer diabetes induced) ,each group was raised 8 weeks In the end of 8th week,to assay the blood glucose(BG) in serum and myocardium of rats from the three groups ; to measure LVSPLVEDP, + dp/dtmax to detect the heart function The left myocardium Sections were made for HE dying and were examined by light microscope Microvascular membrane, and myocardial ultrastructure were studied by electronic microscope. Results :The FBG content levels was increased markedly, + dp/dtmax was decreased in DM group In contrast to DM group. + dp/dtmax Was higher jn TXL group ( P 〈 0.05 ) Under the light microscope demonstrated diabetes group cardiac muscle myofibril arrangement disordered, myo-silk ingredient reduced;under the electron microscope demonstrated cardiac muscle capillaries:endothelial cell obvious swelling, the basilar membrane presented the anomalous obvious accumulation The myocardial cell ultrastructure exceptionally had the Improvement in TXL group. Microvascular membrane, proliferation of the collegen in the extracellular matrix and myocardial ultrastructure were bettered thanDM group. Conclusion : Therearethe abnormalfunctionsofheartandthe abnormalmyocardial ultrastructurein diabetic rats. TXL can inhibittheprogress ofdiabetic cardiomyopathy.
