摘要 RNA干扰(RNA Interference,RNAi)是一种转录后水平的基因沉默(Post-transcriptional Gene Silencing,PTGS)现象,是通过体外人工合成的或体内的双链RNA(Dubble-stranded RNA,dsRNA)在细胞内特异性的降解其同源mRNA,使相应的基因沉默,达到阻止基因表达的目的。因其具有特异、高效、易于操作等优点,为细胞内基因表达研究提供了新思路、新方法,从而使其具有广阔的应用前景。就RNAi机制及其应用研究进行了综述。 RNA interference(RNAi)is a kind of post-transcriptional gene silencing (PIGS) phenomenon. It degrades its homologous mRNA specifically in cell through extracorporeal synthetic or intracorporal double-stranded RNA (dsRNA) to make the corresponding gene silencing and obtain the aim of stopping the gene expression. Because of its advantages such as being efficient, specific and simple and so on, RNAi provided new thoughts and methods for intracell-lar gene expression research. So RNAi will be used widely in the future. The mechanism and the application researches of RNAi were summarized.
