2005年7~8月,在喀什地区采集蚊虫进行病毒分离,从当地居民家畜圈采集的一组库蚊分离到0507JS32病毒,该病毒对C6/36细胞致病变,而对Vero细胞不致病变。电镜观察显示,完整病毒颗粒呈球形,直径55nm,无包膜,衣壳表面壳粒结构明显。基因组核酸电泳显示基因组为12节段双链RNA(double stranded RNA,dsRNA)。利用辽宁病毒(Liaoning virus,LNV)第12基因片段特异性引物对病毒RNA进行RT-PCR扩增,PCR产物进行序列测定,所得核酸序列进行BLAST比较,结果显示,与LNV病毒第12基因片段核酸序列同源性大于89%,证实该病毒为LNV病毒。
Mosquitoes were collected for virus isolation in Kashi from July to August 2005. 0507JS32 virus was isolated from a pool of Culex sp. collected in barn, which could cause cytopathic effects (CPEs) on C6/36 cells but not Vero cells. Viral particles had no envelope and appeared round with diameter of about 55 nm. Capsomeres on the surface of capsid were clearly visible. Electrophoresis of viral genome showed a profile of 1:2 dsRNA segments. Specific primers of twelfth genomic segment of LNV were used for RT-PCR of viral RNA. The sequences of PCR products were compared by BLAST and showed similarity of over 89% with the sequences of LNV in GeneBank. So 0507JS32 virus was identified as LNV.
