摘要
目的:探讨Skp2、p27、p63和COX-2在宫颈鳞状细胞癌及癌前病变中的表达及意义。方法:采用组织微阵技术和SP免疫组化染色方法。结果:标本77例,其中正常宫颈标本14例,CINI-CINⅡ11例,CINⅢ12例,宫颈鳞状细胞癌40例,其中宫颈癌按细胞分化程度分为:I级8例,Ⅱ级23例,Ⅲ级9例。CINI-CINⅡ,CINⅢ,宫颈鳞状细胞癌三组比较,Skp2表达随着宫颈病变的发展而增强,有显著性差异(Hc=26.664,P<0.05),p27表达随着宫颈病变的发展而减弱,有显著性差异(Hc=15.731,P<0.05)。p63表达随着宫颈病变的发展而增强,有显著性差异(Hc=33.935,P<0.05),COX-2表达随着宫颈病变的发展而增强,有显著性差异(Hc=32.408,P<0.05)。Skp2、p63和COX-2在细胞各分化类型(Ⅰ级,Ⅱ级,Ⅲ级)中的表达无显著性差异(P>0.05)。p27均为阳性表达。结论:Skp2和p27为细胞周期的正、负调控因子,其调控的失调,对宫颈癌形成有促进作用。p63表达于具有高度潜能的基底细胞,从CIN发展至癌,p63的表达显著增强,p63阳性表达与p27的阴性表达是否可作为宫颈鳞状细胞癌诊断的指标有待于进一步证实。
Objective: To study the manifestations and significance of Skp2, P27, p63, and COX -2 in squamous cell carcino ma of uterine cervix and its precancerous lesions. Methods: Organization microarray technology and immunity group staining method were used. Results: Among 77 specimens of squamous cell carcinoma of uterine cervix and its precancerous lesions, 14 cases were healthy ones, 11 cases were CIN Ⅰ - CIN Ⅱ , 12 cases were CIN Ⅲ, 40 cases were squamous ceil carcinoma of uterine cervix. According to the differentiation of uterine cervix cancer, 8 cases were the first degree, 23 cases were the second degree, 9 cases were the third degree. . In the comparison of CIN Ⅰ - CIN Ⅱgroup, CIN Ⅲ group and squamous cell carcinoma of uterine cervix group, the martifestation of Skp2 increased with the development of precancerous lesions of uterine cervix, the differences were significant ( Hc = 26. 664, P 〈 0. 05 ) . In contrast, the manifestation of P27 decreased with the development of p lesions of uterine cervix, the differences were significant (He = 15. 731, P 〈 0.05) . The manifestation of p63 increased with the development of precancerous lesions of uterine cervix, the differences were significant ( Hc = 33. 935, P 〈 0. 05) . The manifestation of Skp2, P 63 and COX - 2 didnt differentiate significantly in the three cell differentiation groups (P 〉 0. 05) . All p27 staining given positive manifestation. Conclusion: Skp2 and p27, as the positive and negative cell cycle modulators, contribute to the formation of uterine cervix cancer when their modulation mechanism is in disorder, p63 manifest itself in basal cells with high potential energy, p63 manifestation increasing significantly as C1N develops into cancer. We need further verification to take P63 positive manifestation and p27 negative manifestation as the diagnostic indicator for squamous cell carcinoma of uterine cervix.
