摘要
利用TCA/丙酮沉淀法、直接提取法和两种改良的Tris-HCl提取法对苹果实生树的韧皮部提取蛋白质,并用SDS-PAGE和银染显色的方法对提取的蛋白质结果进行比较,发现:用经过浓缩的的Tris-HCl改良法提取的蛋白质条带清晰,数量多,方法简单且杂质干扰少。因此浓缩的Tris-HCl改良法是适用于苹果韧皮部蛋白质提取的最佳方法。
In the paper, acetone precipitation method (TCA), direct extraction method, two improved Tris-HCl extraction methods are used to extract proteins in phloem of apple seedling trees, SDS-PAGE and silver staining method are used to compare the results of protein extraction, the electrophoresis pattern showed that the protein of each track lane was clear and more, this method was easy,rapid and very few impurity disturbances.Therefore, concentrated and improved Tris-HCl extraction method is the optimum method to extract proteins in phloem of apple.
