HPLC法测定杏香兔耳风中绿原酸和3,5-O-二咖啡酰基奎宁酸的含量 被引量:5

摘要 目的:建立杏香兔耳风药材中绿原酸和3,5-O-二咖啡酰基奎宁酸的含量测定方法。方法:采用HPLC法,使用Dis-covery C18柱(250mm×4.6mm,5μm),以乙腈-0.05%甲酸水溶液为流动相,流速1.0mL.min^-1,梯度洗脱,检测波长为328nm。结果:绿原酸进样量在0.60~3.00μg范围内呈线性关系(r=0.9996,n=5),平均回收率为100.8%;3,5-O-二咖啡酰基奎宁酸进样量在1.30~6.52μg范围内呈线性关系(r=0.9997,n=5),平均回收率为99.4%。结论:本方法操作简便、可靠,重复性好,专属性强,可作为杏香兔耳风药材质量控制方法。 Objective:To establish an HPLC method for determination chlorogenic acid and 3,5-O-dicaffeoylquinic acid in Ainsliaea fragrans Champ.Method:The chromatographic column was Discovery C18(250 mm×4.6 mm,5 μm),UV detection wavelength was 328 nm,a mixture of 0.05% formic acid-water and acetonitrile was used as mobile phase with gradient elution and flow rate was 1.0 mL·min-1.Results:Chlorogenic acid at the quality of 0.60-3.00 μg showed good linearity,the average recovery(n = 5)was 100.8% ;3,5-O-dicaffeoylquinic acid at the quality of 1.30-6.52 μg showed good linearity,the average recovery(n=5)was 99.4%.Conclusion:This methods is fast,reliable and accurate,and may be as one of index used to judge the quality of Ainsliaea fragrans Champ.
