转rolB/pttGA20ox双价基因提高银白杨生根能力及生长速度的研究 被引量:8

摘要 为提高银白杨的扦插生根能力和生长速度,该文利用含有rolB和pttGA20 ox双价基因的植物表达载体对银白杨进行遗传转化,经过PCR和Southern杂交检测,获得了4株转化双价基因的植株。组培苗研究结果表明:转双价基因银白杨在形态特征方面具有rolB和pttGA20 ox单个基因在其他植物中表达的优良性状;在生根能力方面,转化植株的生根时间较对照提前3 d左右,而且生根的数量和长度明显高于对照组银白杨;在生长速度方面,转双价基因银白杨的生长率明显大于对照组银白杨,其生长差异达到了极显著的水平。 To improve cutting rooting ability and growth rate,rolB gene and pttGA20ox gene were transferred into Populus alba.Four transgenic clones were obtained and named GR1-GR4.PCR and Southern analyses confirmed that all the transgenic clones contained those two genes.The in vitro results showed that the morphology characters of all the transgenic clones combined the good traits triggered by the expression of rolB and pttGA20ox genes in other plants respectively.On rooting ability,the root initiation time of transgenic clones was about 3 days ahead of control,the number and length of the roots were also greatly surpassed the control.On height growth,the growth rate of transgenic clones was much fast than that of control and the significance of difference reached extreme level.
