小麦WVp-1基因表达载体的Gateway技术构建及其遗传转化 被引量:1

摘要 为了利用中国特有的抗穗发芽小麦遗传资源,并深入分析Viviparous-1(Vp-1)基因在小麦穗发芽抗性中的作用,从抗穗发芽小麦品种西农979的干种子中分离了Vp-1基因的同源基因,命名为WVp-1。利用Gate-way技术,以植物表达载体pLeela为基础,成功构建了含有双35S启动子的高效表达载体,并转化拟南芥突变体abi3-4,获得了8株阳性植株。 Pre-harvest sprouting (PHS) of grain while it is still in the ear, is due to an early disruption of seed dormancy. PHS reduces the quality of wheat and the value of the grain. Viviparousl (Vp-1) homologues play an important role in seed maturation, dormancy and desiccation. In this study, to make use of genetic resource of pre-harvest sprouting tolerance in Chinese wheats, PCR-based approach was employed to isolate a Vp-1 homologous cDNA clone from Xinong 979, and designated as WVp-1. Using Gateway cloning technology, the plant expression vector of WVp-1 gene driven by the double 35S promoter was successfully constructed and induced into Arabidopsis mutant abi3-4. 8 plants with resistance to Bialaphos were obtain. This laid a basis for the function analysis of WVp-1 gene.
