摘要
JAK2作为JAK家族的一个成员,有JH1~JH7七个结构域,分为FERM区(JH3~JH7)、V617F假激酶区(JH2)和激酶区(JH1)3个部分.JAK2与STATs所构成的信号途径在肾小管上皮细胞转分化、骨髓增殖性疾病的发生、白血病的发生和创伤脓毒症等方面都有重要的影响.但是其具体的作用机理还有待进一步的研究.AG490等JAK2特异性抑制剂和姜黄素等非特异性抑制剂能有效地调节JAK2的异常活性,控制与JAKs/STATs信号途径相关的疾病的发生.
JAK2 is a member of JAK family, it has seven structural domains: JH1 to JH7. These domains are also taken into FERM section (JH3-JH7), V617F bogus kinase section (JH2) and kinase section (JH1). JAK2-STATS signaling pathway has the influences on the transdifferentiation of renal tubular epithelial cells, the taking place of MPDs (myeloproliferative disorder), leucocythemia, sepsis and so on. But more experiments were required to elucidate the mechanism of these phenomena. Specific (AG490) and non specific (curcumin) inhibitor of JAK2 can regulate the activity of JAK2, and control the happening of the diseases related with JAK2.
