摘要
本研究直接从浓缩的鸡胚尿囊液中提取H14禽流感病毒的RNA,并根据已发表的A型流感病毒株的核苷酸序列,设计一对引物,采用反转录-聚合酶链反应(RT-PCR)技术成功地扩增了AIV的核蛋白(NP)基因,以pUC119为载体,以大肠杆菌JM83为受体菌,并通过限制性分析证实。
The present paper describes the nucleoprotein(NP) encoding cDNA of H14 subtype avian influenza virus(AIV)was amplified and cloned.The virus RNA was directly extracted and purified from concentrated chicken embryo allantoic fluid.According to published gene sequence,a pair of specific primers to the NP gene was designed and synthesizde.After that,the NP cDNA was amplified by reverse transcription polymerase chain reaction(RT PCR)and subsequently cloned in pUC119.The presentation of the NP cDNA in transformed E.coli JM83 cells was confirmed by PCR and restriction analysis.
