过氧化苯甲酰-硫胺素荧光光度法的研究与应用 被引量:2

摘要 目的:利用在碱性溶液中硫胺素(VB1)与过氧化苯甲酰(BPO)的荧光反应,建立一种荧光光度法测定药品中硫胺素的新方法。方法:在碱性条件下,BPO将硫胺素氧化成具有较强荧光的硫胺荧,用荧光分光光度计(λex=371 nm,λem=440 nm)测定溶液荧光强度F值,与试剂空白溶液F0比较,以ΔF对硫胺素浓度进行线性回归。同时对体系的反应条件进行了一系列的研究。结果:硫胺素浓度在0.80×10^-5~0.64×10^-4mol/L范围内,体系荧光强度变化(ΔF)与其呈现良好线性关系,线性回归方程为:ΔF=39.768C(×10^-4mol/L)+0.4117,检出限为0.24×10^-5mol/L。对11个浓度为0.4000×10^-4mol/L的硫胺素溶液进行测定,相对标准偏差为1.43%。应用于药物中硫胺素的测定,回收率为94.2%~103.8%。结论:此方法简便、快速,可用于药品中硫胺素的测定。 Objective:To establish a new method for determining thiamine based on the fluorimetric reaction of thiamine with benzoic peroxide in alkaline solutions.Methods:The method was based on the fluorimetric reaction of thiamine with benzoic peroxide in alkaline solutions.And under optimal conditions,the enhanced fluorescence intensity(ΔF=F-F0) had good linear correlation with the concentration of thiamine at 371 nm(λex) and 440 nm(λem).Results:The results showed that the determination range was between 0.80×10^-5 and 0.64×10^-4 mol/L,the linear regression equation ΔF=39.768C(×10^-4mol/L)+0.4117 and the detection limit 0.24×10^-5 mol/L.And the relative standard for 0.4000×10^-4 mol/L thiamine was 1.43%(n=11).The method can be applied to the determination of thiamine in commercial pharmaceutical preparations and the recoveries were between 94.2% and 103.8%.Conclusion:The method of determining thiamine is simple,rapid and accurate.
