摘要
探讨牛磺鹅去氧胆酸(taurochenodeoxycholic acid,TCDCA)的抗炎和免疫调节作用机理。采用实时定量RT-PCR技术检测正常小鼠和免疫抑制模型小鼠脾脏内白介素-2(IL-2)基因mRNA水平表达量的变化。高剂量TCDCA组(0.2g/kg体重)和低剂量TCDCA组(0.1g/kg体重)小鼠脾脏组织内IL-2基因mRNA水平的表达量极显著地高于蒸馏水组(P<0.01);免疫抑制模型高剂量TCDCA和低剂量TCDCA组的小鼠脾脏组织内IL-2基因mRNA水平的表达量均极显著(P<0.01)高于免疫抑制模型,且免疫抑制模型低剂量TCDCA组显著(P<0.05)高于蒸馏水组。TCDCA的抗炎和免疫调节作用与其提高IL-2基因mRNA水平的表达量有关。
To investigate the mechanism of action of anti-inflammatory and immunological regulation, the change of interleukin-2 gene in the level of mRNA in the spleen of healthy and immunosuppressive mouse was detected with the technology of real time RT-PCR. Compared to the control group, the gene expression of interleukin-2 in the spleen of mouse both high dose (0.20 g/ kg body weight) and low does TCDCA (0. 10 g/kg body weight) group could raised (P〈 0.01). Immunosuppressive model groups, high and low TCDCA could markedly revive the gene expression of interleukin-2 inhibited by ciclosporin A and the low TCDCA group markedly toped the control group. The mechanism of anti-inflammatory and immunoregulation has an intimate relationship with enhancing the gene expression of interleukin-2 inhibited in the level of mRNA in the spleen of mouse.
